چکیده
Abstract Study question Do sperm mitochondrial DNA copy number (mtDNAcn) and function correlate with fertilization rates after intracytoplasmic injection (ICSI)? Summary answer We reveal a negative correlation between mtDNAcn ICSI, while higher corresponded to an increase in capacity. What is known already Over 40% of male-factor infertility unknown origin (unexplained idiopathic nature). As current diagnoses remain largely descriptive, the identification markers predicting capacity will hold key towards improved treatment. Mitochondria are fundamental for supporting spermatogenesis fertilization. A recent study showed that altered levels proteins led oocyte-activation deficiency, resulting failure ICSI. Poor semen parameters have also been linked high mtDNAcn, however relationship remains unexplored. Further characterization may uncover prognostic associated sperm-related failure. design, size, duration Twenty normozoospermic samples were included study. These obtained from oocyte donation ICSI cycles, performed March 2021 November 2022. Both fresh cryopreserved analysed. Five million collected ejaculate genomic (gDNA) isolation quantification, remainder sample was used functional analysis. analyses on washed samples, swim-up. Failed fertilization after clinical intracytoplasmic sperm injection.
Intracytoplasmic sperm injection (ICSI) has resulted in pregnancy and birth for many couples, including those with severe male factor infertility. However, even after ICSI, complete failure of fertilization occurs in 1-3% of cycles. Most cases occur due to low number of mature oocytes, failure of oocyte activation or non-availability of appropriate spermatozoa for injection. Given the significa...
متن کاملIn vitro fertilization by intracytoplasmic sperm injection.
Intracytoplasmic sperm injection (ICSI) is the latest, and by far the most efficient, variant of micromanipulation-assisted fertilization, whereby a single spermatozoon is selected, aspirated into a microinjection needle and injected to the oocyte cytoplasm. The development of this technique is mainly linked to application in human assisted reproduction for which it enables fertilization with d...
متن کاملSperm defect severity rather than sperm Source is associated with lower fertilization rates after intracytoplasmic sperm injection.
OBJECTIVE To evaluate the impact of sperm defect severity and the type of azoospermia on the outcomes of intracytoplasmic sperm injection (ICSI). MATERIALS AND METHODS This study included 313 ICSI cycles that were divided into two major groups according to the source of spermatozoa used for ICSI: 1) Ejaculated (group 1; n = 220) and 2) Testicular/Epididymal (group 2; n = 93). Group 1 was subd...
متن کاملIntracytoplasmic Sperm Injection – Factors Affecting Fertilization
The ICSI has become method of choice to achieve fertilization. Fertilization is possible in cases in which the sperm motility and ability to penetrate the zona pellucida are impaired. Injection is possible with sperm obtained from ejaculation, microsurgical epididymal sperm aspiration (MESA), percutaneous epididymal sperm aspiration (PESA), or testicular sperm extraction (TESE). In addition, in...
متن کاملAggressive sperm immobilization prior to intracytoplasmic sperm injection with immature spermatozoa improves fertilization and pregnancy rates.
This study was conducted to determine whether the mode of sperm immobilization prior to intracytoplasmic sperm injection (ICSI) influences fertilization by immature spermatozoa. Of the 837 ICSI cycles evaluated, 81 were performed with epididymal or testicular spermatozoa; 35 cycles with epididymal spermatozoa immobilized in the standard fashion resulted in fertilization and pregnancy rates of 4...
